Gender selection is the process of identifying the sex of embryos through genetic testing (PGD – Preimplantation Genetic Diagnosis) before transfer, and selecting the embryo of the desired sex.
Through this analysis, numerical and structural abnormalities of chromosomes 13, 18, 21, X, and Y are screened. This ensures both the chromosomal health and the sex of the embryo to be transferred. Patients can choose a male or female embryo while being assured that it is not affected by Patau, Edwards, Down syndrome, Turner syndrome, or Klinefelter syndrome.
This increases the likelihood of implantation and eliminates the need for further genetic tests during pregnancy, such as the double test or NIPD.
The procedure starts on day 5 of embryo development, when a biopsy is taken from the embryo’s outer layer (trophectoderm). The biopsy is analyzed in the genetics laboratory, and results are ready within 3–4 hours. Based on the results, the suitable embryo is selected and transferred the same day.

1. Liquefaction:
Semen sample is allowed to liquefy at room temperature or 37°C for 20–30 minutes.
2. Centrifugation (optional):
In the direct swim-Up method, semen is placed directly under the culture medium without centrifugation. In the indirect swim-Up method, the semen is diluted and centrifuged to form a pellet of sperm cells.
3. Overlay with Medium
Culture medium (e.g., Human Tubal Fluid or HEPES-buffered medium) is gently layered on top of the semen or sperm pellet.
4. Incubation
The tube is incubated (typically 30–60 minutes at 37°C) at an angle to encourage sperm to swim upward into the medium.
5. Harvesting
The upper layer of the medium, now enriched with motile sperm, is carefully removed for use in ART procedures.
